Negredo made his senior team debut with Rayo Vallecano in 2005. Playing in the reserve side for most of the time, he impressed several scouts and coaches with his high-scoring ability, which ultimately saw him move to Real Madrid Castilla. He was a part of the squad regularly and started to gain momentum scoring 18 league goals that season. 2007 saw him joining La Liga side UD Almeria where he made his debut against Deportivo de La Coruna. Scoring several crucial goals for the club, he was seen as a great striker having unlimited potential.

Negredo was further moved back to Real Madrid but given the star power of the team in 2009, it was difficult for him to shine. He decided to move to Sevilla in 2009 and made his debut for the club against Valencia.

His campaign with Sevilla gained him worldwide recognition as he made staggering goals in all competitions for the team, earning huge respect and fan following throughout his tenure with the club. After spending numerous times in Sevilla, he joined Premier League side Manchester City in 2013. As he was a part of the Premier League-winning side, he impressed everyone with his knack for scoring goals but was looking dull in the latter stages of the league, scoring less efficiently.

2014 saw him joining Valencia on loan with an option to buy at the end of the season. After having decent performances for the club, Negredo joined the club permanently and played for Valencia till 2016 before returning to the Premier League to play for Middlesbrough. After having another mediocre stint in England, Negredo made his way to Turkey to play for Beşiktaş in 2017. After playing for the Turkish side for over a year, he moved to Al Nasr SC in 2018.

After having another underwhelming tenure at Beşiktaş, Negredo moved to Cádiz in 2020 where he tried to show everyone once again that he has a lot left in him. However, he was released from the club in 2024 after another underwhelming stint, which saw him ultimately moving to Real Valladolid in 2024.
